Tumor cell migration, the basis for metastatic dissemination, is an adaptive process which depends upon coordinated cell interaction with the environment, influencing cell—matrix and cell—cell adhesion, cytoskeletal dynamics and extracellular matrix remodeling. Growth factors and cytokines, released within the reactive tumor microenvironment and their intracellular effector signals strongly impact mechanocoupling functions in tumor cells and thereby control the mode and extent of tumor invasion, including collective and single-cell migration and their interconversions.

Tumorigenesis is a complicated and multistep process, in which successive mutations in oncogenes and tumor-suppressor genes virtually result in enhanced proliferation and resistance to cell death.

A comprehensive understanding of BM structure and mechanics and diverse modes of BM invasion may yield new strategies for blocking cancer progression and metastasis.

The process of tumour-cell invasion and metastasis is conventionally understood as the migration of individual cells that detach from the primary tumour, enter lymphatic vessels or the bloodstream and seed in distant organs. Novel imaging techniques both in vitro and in vivo , together with re-evaluation of histopathological pattern formation in tumours, have provided a detailed view of cellular and molecular migration dynamics in cancer cells. Cancer cells disseminate from the primary tumour either as individual cells, using amoeboid- or mesenchymal-type movement, or as cell sheets, strands and clusters using collective migration.

Cancer invasion and the ability of malignant tumor cells for directed migration and metastasis have remained a focus of research for many years. Numerous studies have confirmed the existence of two main patterns of cancer cell invasion: collective cell migration and individual cell migration, by which tumor cells overcome barriers of the extracellular matrix and spread into surrounding tissues. Two types of migrating tumor cells, mesenchymal fibroblast-like and amoeboid, are observed in each pattern of cancer cell invasion. This review describes the key differences between the variants of cancer cell migration, the role of epithelial-mesenchymal, collective-amoeboid, mesenchymal-amoeboid, and amoeboid- mesenchymal transitions, as well as the significance of different tumor factors and stromal molecules in tumor invasion.
